Iovance Shares Sink: Did the Lung Cancer Update Alter the Outlook?
Iovance Biotherapeutics (NASDAQ:IOVA) shares are trading 35.35% lower at $9.57 on Thursday, October 29, 2026. That same day, the company issued updated results from its IOV-LUN-202 trial of lifileucel in second-line metastatic non-small-cell lung cancer and introduced a precision approach built on tumor biomarkers. The release does not address the share move.
Iovance develops tumor-infiltrating lymphocyte cell therapies for solid tumors. Its marketed product, Amtagvi, is approved in three markets, and the pipeline includes gene-edited and IL-12 tethered versions of the platform.
IOV-LUN-202 Results: How Did Response Rates Vary by Biomarker?
Among 69 non-squamous patients, with a data cutoff of October 14, 2026, the overall response rate was 24.6%, or 17 patients, made up of 15 confirmed responses and two unconfirmed partial responses. An earlier cut of 39 patients showed 25.6%. Independent review is still in progress.
Patients with KRAS wild-type tumors responded at 43.3% (13 of 30), including both complete responses, while KRAS-mutant patients responded at 11.4% (4 of 35). Adding HER2 wild-type status raised the rate to 52.0% (13 of 25), and to 59.1% (13 of 22) among current or former smokers, a group that made up 31.9% of patients but 76.5% of responses.
In a frontline cohort pairing lifileucel with pembrolizumab and no chemotherapy, 6 of 7 patients responded, or 85.7%. The release calls the biomarker findings retrospective and gives no figures for duration of response, progression-free survival or overall survival.
Iovance Development Path: What Do POSITILITY and OPTILISM Involve?
POSITILITY is a prospective, biomarker-enriched cohort inside IOV-LUN-202, with enrollment opening in November 2026 at the first of more than 70 sites. OPTILISM is a planned frontline trial in biomarker-enriched patients, due to start in 2027. The company puts the enriched group at about 45% of U.S. non-squamous cases.
A supplemental application for lifileucel in non-squamous lung cancer is planned for 2027, with about 80 patients expected in the final dataset. Talks with the FDA continue, and a registrational path depends on its feedback. A reduced-dose lymphodepletion regimen is said to have improved safety.
Iovance Sales Momentum: Where Do Revenue and Guidance Stand?
On September 29, 2026, the company increased full-year revenue guidance to $410 million to $420 million from $350 million to $370 million, a rise of $55 million at the midpoint, or about 15%. Second-quarter product revenue was a record $99.3 million, and the treatment center network was near 100, with at least 110 targeted by year-end.
In October, the company also presented ESMO data showing a 62.5% confirmed response rate and a 29.2% complete response rate for lifileucel plus pembrolizumab in frontline melanoma, and a 50% confirmed response rate in soft tissue sarcoma. With 452.97 million shares outstanding, today's price implies a market value near $4.3 billion (calculated).
What to Watch Next: Third-Quarter Earnings on November 5
Iovance will announce third-quarter 2026 results on Thursday, November 5, 2026, giving investors a chance to compare actual revenue with the raised full-year range. The POSITILITY cohort starts enrolling in November, and FDA feedback on the lung cancer pathway ahead of the 2027 supplemental filing is the main regulatory item.
