Tango Therapeutics, Inc. (NASDAQ: TNGX) shares closed at USD 23.34 on October 23, 2026, down 16.43%. The oncology drug developer reported initial vopimetostat combination data and completed a USD 690.0 million public offering in June, then filed its second quarter 2026 report on August 11.
Tango is a Boston, Massachusetts company incorporated in Delaware in May 2020. It aims to discover targeted oncology therapies for the genetic alterations that drive cancer. Its clinical focus is two MTAP-deleted selective PRMT5 inhibitors: vopimetostat (TNG462) for non-CNS cancers, alone and with RAS inhibitors, and TNG456, a brain-penetrant inhibitor for central nervous system cancers including glioblastoma. It has no product approved for sale, and its fiscal year ends December 31.
Second Quarter Loss and Expenses
Tango recognized no collaboration revenue in the second quarter of 2026, compared with USD 3.2 million a year earlier, because the research portion of its Gilead agreement concluded in August 2025. Research and development expense was USD 37.2 million against USD 32.8 million, as spending on vopimetostat and TNG456 increased, partly offset by the end of the TNG908 and TNG260 programs.
General and administrative expense was USD 22.6 million, compared with USD 11.3 million, mainly on higher personnel costs, including share-based compensation. The net loss was USD 55.3 million, or USD 0.37 per share, against USD 38.9 million, or USD 0.35. For six months, the net loss was USD 100.9 million, or USD 0.68 per share, against USD 78.7 million, or USD 0.71. Interest income was USD 3.7 million against USD 1.2 million, and the accumulated deficit was USD 704.0 million.
Combination Data and Pipeline Changes
In June, Tango reported initial data from its Phase 1/2 trial of vopimetostat with Revolution Medicines' RAS(ON) inhibitors in MTAP-deleted, RAS-mutant cancers. With daraxonrasib, the objective response rate in pancreatic cancer was 92%, or 11 of 12 patients, with a 90% six-month progression-free survival rate. In three evaluable lung cancer patients, all three responded.
With zoldonrasib, 27 evaluable patients showed a 52% response rate and 74% six-month progression-free survival. The company described both combinations as generally well tolerated. Grade three events with daraxonrasib included thrombocytopenia, acneiform rash, stomatitis or mucositis and fatigue, and there were no related grade four or five events.
Phase 3 Plans and Programs
Tango intends to move the daraxonrasib combination into Phase 3 for MTAP-deleted pancreatic cancer, subject to regulatory feedback. It plans to share combination data at the European Society for Medical Oncology Congress, held October 23 to 27, 2026. It is deprioritizing TNG260 and shutting down that trial, and it has no current plans to advance TNG961.
Offering and Funding
The June offering comprised 21,166,676 shares at USD 30.00, including 3,000,009 shares from the underwriters' option, plus pre-funded warrants for 1,833,395 shares at USD 29.999. Gross proceeds were USD 690.0 million.
Cash, cash equivalents and marketable securities were USD 1.0 billion at June 30, 2026, which Tango states is sufficient to fund operations for at least twelve months. No shares were sold in the quarter under its USD 100.0 million at-the-market agreement. Tango also states that it will need substantial additional funding because it has no product revenue and expects increasing losses.
Planned Milestones
Tango plans to start a Phase 1/2 trial of vopimetostat with Erasca's ERAS-0015 in the second half of 2026, provide initial TNG456 data in the same period and update monotherapy lung cancer data in 2026. These are company plans and remain subject to clinical and regulatory outcomes.
